Summary

• The highly successful original volume, Protein Function: A Practical Approach, has been completely updated and extended to produce this second edition. Nevertheless, the goal remains the same: the present experimental procedures for analysing protein function that are applicable to many proteins and that can be performed in a typical laboratory without the need for specialized equipment.
• The latest techniques are described in step-by-step detail for protein identification by two-dimensional gel electrophoresis, the expression of cloned genes in E. coli, the preparative folding of proteins, ligand-binding studies, ligand blotting, affinity labelling, and chemical modification of proteins. The assay, purification, and analysis of sequence-specific DNA-binding proteins are also extensively covered, as is the use of chemical cross-linking to probe protein function.
